We are happy to announce a new content series: “Building Blocks for Drug Candidates”. Here we will be highlighting selected sets of Building Blocks that can be used as starting materials for analog synthesis of drug candidate molecules.

Please welcome azetidine-3-ol derivatives and 5-cyclopropyl-1,2-oxazole derivatives – analogs for crucial fragments in Cilofexor. It is a potent, non-steroidal, and intestinally biased FXR agonist. This compound is a drug candidate for the treatment of primary sclerosing cholangitis and is currently in Ph. III preclinical trials.

FXR is a receptor that regulates bile acid metabolism and other metabolic processes. Studies of FXR agonists have shown their potential as therapeutic agents for cholestatic liver disease and non-alcoholic steatohepatitis. However, the ideal pharmacological profile remains a subject of discussion, and further research is aimed at finding more differentiated FXR agonists.
