The paper describes the discovery of new highly selective mAChRs M3 inhibitors with nanomolar activity, which can be promising drug candidates for COPD and asthma treatment. The hits were obtained through in silico modeling with further visual analysis. Following pharmacological studies of trifluoromethyl-containing hexahydropyrimidinones and hexahydropyrimidinethiones, several candidates have been identified as promising precursors of a new generation of cholinolytic drugs.
