We are happy to continue our new inspired by Drug Hunter’s “Small Molecules of the Month” series!
Here are two molecules that were recently highlighted by Drug Hunter!
1. 'Basmisanil' is a negative allosteric modulator (NAM) of gamma-aminobutyric acid A receptor alpha 5 (GABAA-α5). It shows high selectivity against α1, α2, and α3 subunit-containing receptors. Now it is in human trials (Phase II) for treating cognitive deficit in Down syndrome.

2. ‘GNE-9815’ is an oral highly selective pan-RAF Type II kinase inhibitor. It contains a unique hinge-binding motif - pyrido[2,3-d]pyridazin-8(7H)-one, that displays subtle kinase selectivity (). ‘GNE-9815’ demonstrates MAPK pathway modulation in the xenograft mouse model in combination with cobimetinib (MEK inhibitor).
