Here are two molecules that were recently highlighted by Drug Hunter!
1. TAK-653 is a positive allosteric modulator of AMPA receptor which shows little agonistic effects and works as an enhancer for synaptic and cognitive function. It bounds to the ligand-binding domain of AMPA-R similar to glutamate and precisely potentiated a glutamate-induced Ca2+ influx in cells.

2. Compound 59 is a brain penetrant, selective inhibitor of the adaptor protein 2-associated kinase 1 (AAK1) that is known as a potential therapeutic target for neuropathic pain. It is an example of SAR optimization of a series of aryl amide-based AAK1 inhibitors that display efficacy in models for persistent and chronic-constriction-injury-induced pain.
