Here are two molecules that were recently highlighted by Drug Hunter!
1. Compound 4f is a promising reversible monoacylglycerol lipase (MAGL) inhibitor that shows potent inhibitory activity in vitro with good oral absorption and BBB penetration in rodents. This molecule is an example of hit optimization from HTS using SBDD based on interesting spiro scaffolds containing cyclobutane and azetidine moieties.

2. MK-8262 is a unique cholesteryl ester transfer protein (CETP) inhibitor with the potential for being the best-in-class molecule. CETP is known as one of the key regulators of the homeostasis of lipid particles, including high-density lipoprotein (HDL) and low-density lipoprotein (LDL) particles, known as biomarkers for reduced coronary heart disease (CHD) risk. Preclinical and clinical evidence of MK-8262 safety and efficacy by means of HDL increase and LDL reduction show this.
