Shape… What can one say about shape? Every little thing got it or at least looks like something else. Shape is extremely important as it influences the chemical properties. 1,3-substituted bicyclo[1.1.1]pentane and para-phenyl motifs result in the similar shape but bicyclo[1.1.1]pentane-containing compounds have lower toxicity and higher solubility compared with the phenyl derivatives. There are other nonclassical phenyl ring bioisosteres, i.e. cubane, bicyclo[2.2.2]octane.
