A close look at recently developed covalent inhibitors
Last year, a review called “Covalent inhibitors: a rational approach to drug discovery” was published by a group of scientists.
This review aims to provide a short historic overview and focus on recently developed covalent inhibitors (2011–2019).We like the fact that the authors talk about both advantages and possible problems of such components in drug discovery.

Fluorine-containing blockbuster drugs
Last year, a group of researchers published a review called “Next generation organofluorine blockbuster drugs”. This work highlights a section of most successful organo-fluorine drugs (eg. sitagliptin (diabetes), sofosbuvir (hepatitis C), emtricitabine (HIV)), their synthesis, and mode of action.

Spirocycles as Bioisosteres for Aromatic Fragments
Last month, a group of researchers published an article on the design and synthesis of novel spiro derivatives as potent and reversible Monoacylglycerol Lipase inhibitors (MAGL).
The authors used a structure-based drug discovery strategy to optimize an aromatic HTS hit to the final compound by replacing the binding site of the molecule with aliphatic moieties.
Based on the article, we prepared a set of spirocyclic amides, that can become a great start for your research.

Sulfonimidamides as bioisosteres in peptide-based drug discovery
Last month, a group of researchers posted an article called ‘Antibacterial sulfonimidamide-based oligopeptides as type I signal peptidase inhibitors: Synthesis and biological evaluation’.
The goal of the study was to determine if changing an amine group to sulfonamide or sulfonimidamide in peptides will result in marked differences in their properties.We caught up the concept and prepared a set of sulfonimidamides for your research!

Oxazolidinone Scaffolds as Antibacterial Drugs
This July, a group of researchers posted an article called ‘Current landscape and future perspective of oxazolidinone scaffolds containing antibacterial drugs’. This work is important because it covers an increasingly serious problem - bacterial resistance to drugs.
The core structure gets modified in order to achieve better molecule parameters and higher activity against bacteria.We at Chemspace prepared a set of compounds based on phenyl oxazolidinone scaffold for you!

This July, Belumosudil (REZUROCK) received approval by the FDA as the treatment of chronic graft versus host disease (cGvHD).
It is an orally available inhibitor of Rho-associated coiled-coil kinase 2 (ROCK 2).
Belumosudil contains quinazoline moiety in its backbone.

Reducing lipophilicity with the help of azaspiro[3.3]heptanes
In 2019, a group of researchers discovered, that azaspiro[3.3]heptanes can act as replacements for morpholines, piperidines, and piperazines in a medicinal chemistry context.
The authors concluded that azaspiro[3.3]heptanes are most likely not suitable as bioisosteres, when not used as terminal groups.
